Research-related questions like the different stages of a clinical trial.  
Some important guidelines relevant to SA patient population, e.g., HIV and TB guidelines. GCP guidelines are important to know well.  
Ethical questions like patient consent.

Any research background/experience you might have.  
Practical questions about how to do research, where to find candidates, how to prioritize the different steps, etc.  
Beyond that, they might try to find out a bit more about who you are, what your commitments and plans are, etc.
